Mysterious lilo error

When I run LILO on my machine, I get the following error:

First boot sector doesn't have a valid LILO signature

Re-running lilo doesn't help, and I don't want to reformat the drive. How can
I wipe the old LILO signature and get this one to work?

Any ideas?
